Yes, you can!

In the Data Settings, under the "Fields" section, simply select the field po_number to include the purchase order number in your Shopify order CSV/Excel export.

Note that this field is only available for Shopify B2B orders.  In the Shopify Admin, this field maps to this section shown below:
